(1) Functional partition
All samples are considered to have biological hazards or potential biological hazards. In order to prevent biological hazards from spreading with the air or aerosol in the sample warehouse or even outside the area, the preservation center has a clean area, a semi-polluted area and a polluted area. The clean area includes the entrance hall, the security monitoring room and the reagent consumables warehouse, and the semi-polluted area includes the refrigerator area, the normal temperature preservation area, the automatic preservation area and the liquid nitrogen preservation area, The contaminated area includes sample receiving and temporary storage area, sample processing and quality control area, and waste temporary storage room; At the same time, the functional areas that directly contact the samples are equipped with full-row biosafety cabinets or fume hoods.
(2) Flow direction setting
Human flow, sample flow, logistics and sewage flow enter the reservoir area from different entrances, human flow enters from the clean area, and material sample flow enters from the polluted area. A striking "biological pollution" sign and relevant system are posted at the entrance of the sample flow to remind the staff to take safety protection measures.
(3) HVAC
The preservation center has strict requirements for the design of laboratory differential pressure. The design of differential pressure between laboratories is conducive to the flow of air from the clean area to the polluted area, and the maximum reduction of indoor backflow and vortex; At the same time, the sample storage equipment has high requirements for the ambient temperature and humidity: the humidity requirement of the automatic storage area is less than 40%, and the industrial dehumidification system must be equipped in the area to ensure that the ambient humidity meets the requirements; The nitrogen generated by the volatilization of liquid nitrogen in the liquid nitrogen storage area has a great impact on the safety of personnel. To ensure the safety of personnel, on the one hand, the indoor oxygen concentration is monitored through the oxygen concentration monitoring, on the other hand, the indoor ventilation times are not less than 6 times per hour. When the nitrogen concentration exceeds the standard, the ventilation times per hour are increased to 12 times.
(4) Standby energy
The samples in the sample library are stored under ultra-low temperature and deep low temperature storage conditions. It is very important to maintain the stable power supply and liquid nitrogen supply of the sample library. In order to ensure the electricity safety of the samples, the sample library is equipped with three sets of UPS (one for 500KVA, 400KVA and 30KVA respectively) and three sets of generator sets (600kw, two for use and one for standby) for the preservation center, which can ensure the uninterrupted power supply of all storage equipment.
(5) Fire safety
The fire alarm smoke monitoring system and fire extinguishing device have been established. Heptafluoropropane will be used in the area where the automatic ultra-low temperature storage equipment, automatic liquid nitrogen tank, automatic pre-treatment and quality control equipment are placed; There are two spray devices in the laboratory area on the first floor, and the pool is equipped with an eye washer.
(6) Building load bearing
The automatic ultra-low temperature storage equipment, automatic gas-phase liquid nitrogen tank, automatic pre-treatment and quality control equipment and other large equipment used in the preservation center have a large dead weight. The bearing capacity of the first floor will increase with the increase of sample storage, and the bearing capacity of the building will reach 1000kg/m2.
(7) Safety monitoring
The access control management module is set for all access channels of the sample library preservation center, which can realize personnel management in different areas by assigning different access control permissions; A video monitoring system is installed in the automatic ultra-low temperature equipment, the automatic liquid nitrogen tank and the public walkway of the building to realize 24-hour real-time monitoring; A temperature and humidity monitoring system is set in the automatic storage area and the liquid nitrogen storage area. When the temperature exceeds 20 ℃ ± 5 ℃ and the humidity is more than 40%, start the ventilation device to adjust the temperature and humidity to ensure that the equipment operates under appropriate temperature and humidity conditions and ensure the stability of sample storage conditions; Oxygen concentration monitoring is installed near the liquid nitrogen area, and the alarm will be started when the oxygen concentration is lower than 18% to ensure the safety of personnel.
