The layout of liquid nitrogen area is shown in Figure 4.
The liquid nitrogen storage area is equipped with two bases and liquid nitrogen pipelines that can install 10m3 liquid nitrogen tower; 2 automatic gas-phase liquid nitrogen storage equipment and 24 conventional gas-phase liquid nitrogen tanks are arranged; At the initial stage, it is planned to purchase 1 automatic gas-phase liquid nitrogen storage equipment and 5 conventional gas-phase liquid nitrogen tanks (realizing 5 for use and 1 for standby), rent a 5m3 liquid nitrogen tower, and prepare relevant equipment according to the actual development of the sample library in the later stage.
(1) Vapor phase liquid nitrogen tank
In order to avoid cross contamination of samples in the tank, the liquid nitrogen storage area uses gas-phase liquid nitrogen tanks to store samples to ensure the biological safety of the sample bank.
The conventional gas-phase liquid nitrogen tank mainly consists of four parts: tank body, pipeline, control system and sample rack. It has the following advantages: ① monitoring and early warning of key information: each gas-phase liquid nitrogen tank is equipped with a temperature and liquid level monitoring system. When the temperature or liquid level exceeds the threshold, it will send a notice to the reservoir management personnel to ensure that the temperature in the tank is stable; ② Improve staff's work efficiency: on the one hand, liquid nitrogen tanks are equipped with automatic filling function. When there is low liquid level in the tank, liquid nitrogen can be filled automatically to stabilize the amount of liquid nitrogen in the tank and ensure that the temperature in the tank is stable. On the other hand, each equipment is equipped with lifting arm to assist the staff in the warehouse to take samples; ③ Improve the diversity of storage specifications: the sample rack placed in the tank can be customized according to the storage requirements.

The automatic gas-phase liquid nitrogen tank adds a tube picking function module on the basis of the configuration of the conventional gas-phase liquid nitrogen tank, which can pick up the single-tube SBS frozen storage pipe, realize the automatic access of the frozen storage pipe and the single-tube/whole box entry and exit functions, and the temperature of the tube picking area is about - 150 ℃ (at this time, the sample temperature is lower than - 135 ℃, which is in the glass state), and the sample stability is better.
The daily operation and monitoring data of the automatic gas-phase liquid nitrogen tank can be synchronously uploaded to the sample library management software and monitoring system, that is, the data exchange is realized at the information management level to facilitate the management of the reservoir area.
(2) Liquid nitrogen support system
Including liquid nitrogen tower and liquid nitrogen pipeline, the system plays the following roles in operation: ① operation safety: the safe operation of liquid nitrogen support system is the key link to be considered in the development of sample library, and the key safety valve and remote control system are set in the system to ensure the normal gas-liquid pressure in the liquid nitrogen tower and pipeline, prevent the occurrence of liquid nitrogen leakage, and ensure the operation safety; ② Maintaining pressure stabilization: the automatic filling of gas-phase liquid nitrogen tank and the liquid nitrogen refrigeration function of automatic ultra-low temperature storage equipment both depend on the pressure stabilization of the liquid nitrogen support system, that is, the liquid nitrogen in the pipeline can be used for supply in real time, and the nitrogen naturally evaporated in the tower can be used to maintain the positive pressure of the liquid nitrogen pipeline system, which can reduce the formation of ice crystals in the pipeline, avoid pipeline blockage, and improve the stability of the liquid nitrogen support system in pressure stabilization operation. ③ The layout of the liquid nitrogen tank is flexible: since the liquid nitrogen tank in this area will be purchased in batches, the design of the liquid nitrogen outlet can meet the liquid nitrogen demand of the tank with a diameter of 1.5 meters and about 1 meter, which is convenient for the reasonable placement of the liquid nitrogen tank according to the tank size in the later stage.
